Effects of dimethyl sulfoxide on the globally ischemic heart: possible general relevance to hypothermic organ preservation.

چکیده
Dimethyl sulfoxide (DMSO) is routinely used to prevent freezethaw-induced damage to a variety of biological systems. In that context, relatively high drug concentrations in excess of one molar are usually used. However, lower concentrations of the drug produce many pharmacological effects (26) which may be efficacious for reducing hypoxiaor ischemia-induced cell damage which is often encountered during organ preservation protocols that do not necessarily involve freezing. During the course of a study of several interventions which might be useful to prevent damage to the globally ischemic heart, we observed a relatively discrete but perhaps important action of dimethyl sulfoxide. When added to hypothermic cardioplegic solutions, 70 mM dimethyl sulfoxide significantly and consistently prevented alterations of mitochondrial oxidative phosphorylating activity after 2 hr of global ischemia and reperfusion. Based on these findings, the known pharmacological spectrum of action of dimethyl sulfoxide, and a postulated mechanism by which it exerted its protective effects, we were led to speculate that the drug might have more widespread applicability to organ preservation than it currently enjoys.

Cardiac pharmacology of dimethyl sulfoxide and its postulated relevance to organ preservation in ischemic or hypoxic states.
Beginning with the studies published by Sams and colleagues' in 1966, there has been a growing list of effects of dimethyl sulfoxide (DMSO) on the myocardium. Many of the studies of cardiac effects of DMSO focused on its use in high (multimolar) concentrations to reduce freeze-thaw-induced injury to the heart, with the ultimate goal being long-term cryopreservation of human hearts for transplan...

The effect of modified vitrification was assessed on cellular development capability in mouse embryos cultured in vitro. In this study, 466 embryos (from zygote to morula stages) were vitrified then thawed embryos have been incubated for in vitro farther development up to blastocyst stage. Also, vitrification and thawing procedures were the same for all experimental groups. Mouse different embr...
متن کاملEffects of Dimethyl Sulfoxide and Mutations on the Folding of Abeta(25-35) Peptide: Molecular Dynamics Simulations
The 25-35 fragment of the amyloid β (Aβ) peptide is a naturally occurring proteolytic by-product of its larger parent molecule that retains the amyloid characteristics and toxicity of the full length parent molecule. Aggregation of this peptide occurs rapidly in aqueous solutions and thus characterization of its folding process is very difficult. In the present study, early stages of Aβ(25–35) ...

متن کاملThe Effect of Hypothermic and Cryogenic Preservation on Engineered Neural Tissue
This study explored different approaches to preserve engineered neural tissue (EngNT), a stabilized, cellular collagen hydrogel containing columns of aligned Schwann cells for nervous system repair. The ability to preserve EngNT without disrupting cellular and extracellular components and structures is important for clinical translation and commercialization. Stabilized cellular gels and EngNT ...
